目的 :研究鲨鱼软骨制剂 ( SCP)对人胃癌细胞 ( MGC80 -3 )、红白血病细胞( K562 )、人脐静脉血管内皮细胞 ( VEC3 4 0 )的生长抑制作用 ,探讨其抗癌作用机制。方法 :采用盐酸胍抽提、丙酮分级沉淀、烘干、微孔滤膜过滤等步骤从鲨鱼软骨粉中提取鲨鱼软骨制剂 ;采用 MTT法检测不同浓度 SCP对体外培养的 MGC80 -3细胞系、K562细胞系和 VEC3 4 0细胞系的生长抑制作用。结果 :SCP明显抑制 MGC80 -3、 K562和 VEC3 4 0三种细胞系的生长 ,其 IC50值分别为 1 mg/ml、 0 .75~ 1 .5mg/ml和 3 mg/ml。结论 :SCP能直接抑制肿瘤细胞生长 ,又能抑制血管内皮细胞的生长 ,具有潜在肿瘤治疗价值
Objective: To study the growth inhibitory effects of shark cartilage preparation (SCP) on human gastric cancer cells (MGC80-3), erythroleukemia cells (K562), and human umbilical vein endothelial cells (VEC304) and to explore its anticancer mechanism. METHODS: Shark cartilage preparations were extracted from shark cartilage powder using guanidine hydrochloride extraction, acetone fractionation, drying and microfiltration membrane filtration. MCT assay was used to detect different concentrations of SCP against MGC80-3 cell line and K562 cultured in vitro. Growth inhibition of cell lines and VEC304 cell lines. RESULTS: SCP significantly inhibited the growth of MGC80-3, K562 and VEC304 cell lines with IC50 values of 1 mg/ml, 0.75 to 1.5 mg/ml and 3 mg/ml, respectively. Conclusion : SCP can directly inhibit the growth of tumor cells and inhibit the growth of vascular endothelial cells. It has potential therapeutic value for tumors.
