目的建立短筒兔耳草中松果菊苷的RP-HPLC测定方法。方法短筒兔耳草经流动相超声提取后,用RP-HPLC法测定。采用Alltech Prevail C18(250mm×4.6mm,5μm)色谱柱,流动相为乙睛-甲醇-1%醋酸(10:15:75),体积流量1.0mL/min,检测波长334 nm,柱温为室温。结果松果菊苷进样量在0.46~18.40μg内与峰面积呈良好的线性关系(r=0.999 9);平均回收率为101.11%,RSD为2.09%(n=6)。结论采用RP-HPLC测定短筒兔耳草药材中松果菊苷的含量,该方法准确、快速、简便,重复性好,适用于短筒兔耳草中松果菊苷的质量控制。
Objective To establish a RP-HPLC method for the determination of echinacosin in Pleurotus ostreatus. Methods Short-eared rabbit ear grass was extracted by mobile phase and determined by RP-HPLC. The mobile phase was acetonitrile-methanol-1% acetic acid (10:15:75), the volume flow rate was 1.0 mL / min, the detection wavelength was 334 nm and the column temperature was room temperature, using Alltech Prevail C18 column (250 mm × 4.6 mm, . Results The elution volume of echinacoside was 0.46 ~ 18.40μg, with a good linear relationship (r = 0.999 9). The average recovery was 101.11% and the RSD was 2.09% (n = 6). Conclusion RP-HPLC was used to determine the content of echinacoside in Elaeagnus angustifolia. The method is accurate, rapid, simple and reproducible. It is suitable for the quality control of Echinacea in Eupatorium.
