本文用氚标胸腺嘧啶核苷掺入DNA合成法测定自发性高血压大鼠(SHR)与正常对照鼠的培养主动脉血管平滑肌细胞(VSMC)增殖,观察L-苯丙氨酸对细胞增殖、细胞生长及原癌基因c-fos、c-myc表达的影响。结果显示:(1)L-苯丙氨酸剂量依赖性地抑制血清、碱性成纤维细胞生长因子及凝血酶诱导的DNA合成;(2)L-苯丙氨酸剂量依赖性地抑制细胞对血清的增殖反应;(3)L-苯丙氨酸抑制血清诱导的c-fos、c-myc的表达;(4)L-酪氨酸,L-组氨酸及D-苯丙氨酸不能产生类似L-苯丙氨酸的抑制作用。这些资料说明L-苯丙氨酸具有直接、特异性的抗VSMC增殖作用,提示L-苯丙氨酸在SHR大鼠中的抗高血压作用可能与血管平滑肌细胞增殖受抑有关。
In this paper, tritiated thymidine incorporation into DNA synthesis was used to measure the proliferation of aortic vascular smooth muscle cells (VSMCs) from spontaneously hypertensive rats (SHR) and normal control mice. The effects of L-phenylalanine on cell proliferation, Cell growth and the expression of proto-oncogene c-fos and c-myc. These data indicate that L-phenylalanine has a direct and specific anti-proliferative effect on VSMC, suggesting that anti-hypertensive effects of L-phenylalanine in SHR rats may be related to inhibition of vascular smooth muscle cell proliferation.
