(一)配方筛选A.马铃薯200g,葡萄糖20g,琼脂20g,KH_2PO_43g,MgSO_41.5g,水1000ml;B.大米250g,葡萄糖20g,琼脂18g,KH_2PO_41.5g,MgSO_40.5g,水1000ml;C.小米200g,葡萄糖20g,琼脂18g,麸皮5g(取汁),KH_2PO_41.5g,MgSO_40.5g,水1000ml;D.小麦250g,葡萄糖20g,琼脂20g,KH_2PO_43g,Mg-SO_41.5g,蛋白胨5g,加水1000ml。pH 值均自然。(二)试验方法取小麦250g,加水1000ml加热煮沸后保持30分钟,用四层纱布过滤,把滤液补足水量至1000ml。煮沸后加入琼脂、葡萄糖、蛋白胨、磷酸二氢钾和硫酸镁,待其全部溶化后趁热分装试管。高压灭菌30分钟。其它配方和小麦取汁相同。每配方处理重复10支试管,每支接入约5mm 见方的菌块,放入25±1℃恒温下培养。

(B) Test Method Take wheat 250g, add water 1000ml boil and keep for 30 minutes, with four gauze filter, the filtrate make up to the water to 1000ml. After boiling add agar, glucose, peptone, potassium dihydrogen phosphate and magnesium sulfate, until it is fully dissolved hot melt dispensing test tube. Autoclave for 30 minutes. Other formula and wheat juice the same. Each formula to deal with repeated 10 tubes, each access about 5 mm square bacteria, 25 ± 1 ℃ constant temperature training.
