目的:观察小檗碱对高脂饮食诱导的小鼠慢性肾脏损伤的影响及肾脏内巨噬细胞的变化。方法:将30只6周龄C57BL/6小鼠随机分为正常对照组、高脂饮食组、小檗碱治疗组,每组10只,检测总胆固醇、甘油三酯、血肌酐、尿素氮、胱抑素C(Cys C)、24 h尿量和尿白蛋白,HE染色观察肾脏形态病理学变化。Western blot检测白细胞介素(IL)-1β和诱导型一氧化氮合酶(iNOS)的表达水平。免疫荧光染色检测巨噬细胞标记物F4/80的表达变化。免疫组化染色观察M1型巨噬细胞标记物iNOS的表达情况。结果:高脂饮食组小鼠血脂、Cys C、24 h尿量和尿白蛋白水平显著升高(P<0.05);IL-1β、F4/80和iNOS在肾脏表达显著增高(P<0.05)。小檗碱治疗组小鼠血脂、Cys C、24 h尿量和尿白蛋白水平有所降低;IL-1β、F4/80和iNOS的表达均较高脂饮食组减少(P<0.05)。结论:小檗碱可以减轻高脂饮食诱导的小鼠慢性肾脏损伤,这种保护作用是通过减少肾脏组织浸润的巨噬细胞,尤其是减少M1型巨噬细胞的浸润而发挥的。
Objective: To observe the effects of berberine on chronic kidney injury induced by high-fat diet and the changes of macrophages in kidneys. Methods: 30 6-week-old C57BL / 6 mice were randomly divided into normal control group, high-fat diet group and berberine group, with 10 rats in each group. Total cholesterol, triglyceride, serum creatinine, Cystatin C (Cys C), 24 h urinary albumin and urine albumin, HE staining were observed morphological changes of the kidney. Western blot was used to detect the expression of interleukin (IL) -1β and inducible nitric oxide synthase (iNOS). The change of macrophage marker F4 / 80 expression was detected by immunofluorescence staining. The expression of iNOS, a marker of macrophage type M1, was observed by immunohistochemistry. Results: The levels of serum lipids, Cys C, 24 h urinary albumin and urinary albumin in high fat diet group were significantly increased (P <0.05). The expressions of IL-1β, F4 / 80 and iNOS in kidney were significantly increased . Compared with the high-fat diet group, the levels of serum lipids, Cys C, 24 h urinary albumin and urinary albumin in berberine-treated mice decreased. The levels of IL-1β, F4 / 80 and iNOS in the mice treated with berberine decreased significantly (P <0.05). Conclusion: Berberine can alleviate chronic kidney injury induced by high-fat diet in mice by reducing the infiltration of macrophages in the kidneys, especially the macrophages.
