利用 ELISA 双抗夹心法测定两组接受庆大霉素(剂量分别为100mg/kg·d 与50mg/kg·d)腹腔注射的大鼠尿中近曲小管刷状缓微绒毛抗原(BBMA)。组织病理学检查显示两组大鼠分别在注射庆大霉索后第2天(100mg 剂量组)和第6天(50mg 剂量组)发生近曲小管上皮细胞变化、坏死与脱屑。与此同时,尿 BBMA 的排出开始明显超过正常对照组(P<0.01;P<0.05)。其变化与电镜所见肾小管损伤的进展程度相符。100mg 剂量组直到第4天后才出现尿 NAG、尿蛋白和尿溶菌酶增高以及肌酐廓清率下降,而这些指标在50mg 剂量组的实验期间均无显著改变。实验结果表明尿BBMA 的测定有助于急性肾小管损伤的早期定位诊断。
The urinary proximal brush tube microtumor microtubule antigen (BBMA) of rats receiving gentamicin (100 mg / kg · d and 50 mg / kg · d, respectively) was measured by ELISA. Histopathological examination showed that proximal tubule epithelial cell changes, necrosis and desquamation occurred on the 2nd day (100mg dose group) and the 6th day (50mg dose group) in the two groups. At the same time, the urinary BBMA excretion started significantly more than the normal control group (P <0.01; P <0.05). The changes consistent with the extent of the progress of renal tubular damage seen by electron microscopy. Urine NAG, urinary protein and urine lysozyme increased, and creatinine clearance rate decreased in the 100 mg group until day 4, and no significant changes were observed in these parameters during the 50 mg dose group. The experimental results show that the determination of urinary BBMA is helpful for the early localization diagnosis of acute tubular injury.
