为了研究金钱草粗提物对大鼠主动脉的舒张作用,初步探讨其作用机制,试验采用大鼠主动脉张力测试的方法,快速分离大鼠主动脉并固定于多通道血管张力测试系统中,用NO合酶抑制剂(L-NAME)观察筛选金钱草提取物对大鼠主动脉的舒张作用。结果表明:金钱草水部位、正丁醇部位、石油醚部位和乙酸乙酯部位粗提物(1 g/L)对大鼠主动脉均有舒张作用,其中乙酸乙酯部位舒张作用最强;乙酸乙酯部位能剂量依赖性地舒张大鼠主动脉,去除内皮细胞后其舒张能力显著降低;预孵育L-NAME能显著抑制乙酸乙酯部位对大鼠主动脉的舒张作用。说明金钱草乙酸乙酯部位能显著舒张大鼠主动脉,其机制可能与其激活内皮细胞一氧化氮合酶、促进内皮细胞释放一氧化氮有关。
In order to study the relaxation effect of the extract of Rumex stellata on the aorta of rats, the mechanism of its action was preliminarily investigated. The rat aortic tension test was used to rapidly separate the aorta and fix it in the multi-channel vascular tension test system. The NO synthase inhibitor (L-NAME) was used to observe the relaxation effect of the extract from Lysimachiae to aorta in rats. The results showed that water fraction, n - butanol fraction, petroleum ether fraction and ethyl acetate extract (1 g / L) had a relaxing effect on the aorta of rats, and the strongest effect of ethyl acetate was observed. Ethyl acetate could relax the aorta in a dose-dependent manner and significantly reduce the diastolic capacity of endothelial cells after removing endothelial cells. Pre-incubation of L-NAME significantly inhibited the relaxation of ethyl acetate fraction on the aorta of rats. The results showed that the ethyl acetate fraction could significantly relax the aorta in rats, which may be related to the activation of endothelial nitric oxide synthase and the release of nitric oxide by endothelial cells.
