目的探讨黄芪多糖对子宫内膜癌患者外周血单核细胞源性树突状细胞(DCs)成熟的影响。方法从子宫内膜癌患者外周血中分离单核细胞,以加入IL-4、GM-CSF树突状细胞专用培养基培养5d后,以不同浓度的黄芪多糖(终浓度分别为0、50mg/L、100mg/L、150mg/L)刺激48h,镜下观察DCs形态变化,采用流式细胞术检测树突状细胞表型CD1α、CD83、CD86的表达,以及ELISA法检测细胞上清液中IL-12的含量。结果 150mg/L APS组细胞生长状态最好,具有典型的树枝状突起;150mg/L APS组、100mg/L APS组DCs表面CD1α、CD83及CD86的表达较对照组、50mg/L APS组明显上调(P<0.05);150mg/L APS组、100mg/L APS组细胞上清液中IL-12含量高于对照组、50mg/L APS组(P<0.05)。结论黄芪多糖可促进子宫内膜癌患者外周血单核细胞源性DCs成熟以及IL-12的分泌。
Objective To investigate the effects of astragalus polysaccharides on the maturation of peripheral blood mononuclear cells derived dendritic cells (DCs) in patients with endometrial cancer. Methods Mononuclear cells were isolated from peripheral blood of patients with endometrial carcinoma and cultured in special medium containing IL-4 and GM-CSF dendritic cells for 5 days. The cells were treated with different concentrations of APS (0, 50 mg / L, 100mg / L, 150mg / L) for 48h. Morphological changes of DCs were observed under microscope. Flow cytometry was used to detect the expression of dendritic cell phenotypes CD1α, CD83 and CD86, -12 content. Results The cells in the 150mg / L APS group had the best growth status with typical dendrites. The expressions of CD1α, CD83 and CD86 on the surface of DCs in 150mg / L APS group and 100mg / L APS group were significantly higher than those in the control group and 50mg / L APS group (P <0.05). The content of IL-12 in supernatant of 150mg / L APS group and 100mg / L APS group was higher than that of control group and 50mg / L APS group (P <0.05). Conclusion Astragalus polysaccharide can promote the maturation of peripheral blood mononuclear cells and secretion of IL-12 in patients with endometrial carcinoma.
