目的:探讨参芪扶正注射液含药血清对化疗药物所造成免疫抑制的减毒作用。方法:选用Wistar雄性大鼠20只,随机分为含药血清组和空白血清组。含药血清组按40 g.kg-1.d-1的剂量尾静脉注射参芪扶正浓缩液,每日给药2次,连续给药5次。空白血清组给予生理盐水代替参芪扶正浓缩液,余相同。2组大鼠于末次给药后1 h后自腹主动脉采血,分离血清。研究设药物组和对照组。药物组分别用5-氟尿嘧啶(5-Fu)注射液(50 mg.L-1)、顺铂注射液(6.25 mg.L-1)、参芪扶正注射液含药血清作用于小鼠巨噬细胞系RAW264.7细胞;对照组用空白血清作用于RAW264.7细胞。利用酶标仪对作用后的RAW264.7细胞进行检测,观察5-Fu注射液、顺铂注射液、含药血清对RAW264.7细胞增殖的影响。结果:单用参芪扶正注射液组与空白对照组相比能明显促进RAW264.7细胞增殖(P<0.01)。5-Fu、顺铂注射液联合含药血清后与单用5-Fu、顺铂注射液相比,对于RAW264.7细胞增殖的抑制明显降低,其差异皆有显著性(P<0.01)。结论:参芪扶正注射液在体外环境下可促进巨噬细胞的增殖,改善化疗药物所造成的免疫抑制,对不同化疗药物的减毒趋势相同而减毒强度不同。
OBJECTIVE: To study the attenuated effect of Shenqi Fuzheng injection-containing serum on the immunosuppression caused by chemotherapy drugs. Methods: Twenty Wistar male rats were randomly divided into drug-containing serum group and blank serum group. Drug-containing serum group by 40 g.kg-1.d-1 dose of tail vein injection of Shenqi Fuzheng concentrated solution, administered twice daily, continuous administration of 5 times. Blank serum group instead of Shenqi Fuzheng concentrated liquid, the same. Two groups of rats after 1 h after the last administration from the abdominal aorta blood, serum was isolated. Study drug group and control group. Drug groups were treated with 5-Fu injection (50 mg.L-1), cisplatin injection (6.25 mg.L-1), Shenqi Fuzheng injection-containing serum on mouse macrophages Cell line RAW264.7 cells; control group with blank serum on RAW264.7 cells. The effects of 5-Fu injection, cisplatin injection and drug-containing serum on the proliferation of RAW264.7 cells were detected by using microplate reader. Results: Compared with the blank control group, Shenqi Fuzheng injection alone could significantly promote the proliferation of RAW264.7 cells (P <0.01). 5-Fu, cisplatin injection combined with serum containing 5-Fu, cisplatin alone compared to the proliferation of RAW264.7 cell proliferation was significantly reduced, the difference was significant (P <0.01). Conclusion: Shenqi Fuzheng injection can promote the proliferation of macrophages in vitro and improve the immunosuppression caused by chemotherapy drugs. The attenuating intensities of different chemotherapeutic drugs are the same but different.
