目的探讨过氧化氢(H2O2)摄入后小鼠睾丸生精细胞环氧化酶-2(COX-2)和Caspase-3表达的影响。方法健康雄性昆明种小鼠30只,随机分为对照组10只和实验组20只。对照组小鼠喂食自来水,实验组小鼠以0.3 g.L-1H2O2(0.3 g.L-1剂量组)和3.0 g.L-1H2O2(3.0 g.L-1剂量组)溶液喂食。30 d时处死各组小鼠,取出其睾丸,免疫组织化学链霉亲和素-生物素酶复合物染色法检测其生精细胞COX-2和Caspase-3表达的变化。结果与对照组比较,0.3 g.L-1剂量组和3.0 g.L-1剂量组COX-2、Caspase-3表达均显著升高,差异有统计学意义(Pa<0.05,0.01),与0.3 g.L-1剂量组比较,3.0 g.L-1剂量组COX-2无显著升高,Caspase-3显著升高(P<0.01)。结论 HO经口摄入可造成小鼠睾丸生精细胞的损伤。
Objective To investigate the effects of hydrogen peroxide (H2O2) on the expression of COX-2 and Caspase-3 in mouse testis. Methods Thirty male Kunming mice were randomly divided into control group (n = 10) and experimental group (n = 20). The control mice were fed tap water. The mice in the experimental group were fed with 0.3 g · L-1H2O2 (0.3 g · L-1 dose) and 3.0 g · L-1H2O2 (3.0 g · L-1 dose). The mice were sacrificed on day 30 and their testis was removed. The expression of COX-2 and Caspase-3 in spermatogenic cells was detected by immunohistochemical streptavidin-biotin complex staining. Results Compared with the control group, the expressions of COX-2 and Caspase-3 in 0.3 gL-1 and 3.0 gL-1 groups were significantly increased (P <0.05, 0.01) Compared with the dose group, COX-2 was not significantly increased and Caspase-3 was significantly increased in the 3.0 gL-1 dose group (P <0.01). Conclusions Oral intake of HO can cause the damage of spermatogenic cells in mouse testis.
