目的:研究西藏绿萝花提取物对α-葡萄糖苷酶的抑制及抗氧化作用。方法:将西藏绿萝花的70%乙醇提取物依次用石油醚、乙酸乙酯、正丁醇萃取,然后测定各部分对α-葡萄糖苷酶的抑制及抗氧化作用。结果:乙酸乙酯的萃取部分对α-葡萄糖苷酶的活性抑制较强,半数抑制浓度IC50为43.63μg/mL,比阿卡波糖抑制α-葡萄糖苷酶的IC50值250μg/mL低的多,抵制动力学表明为竞争性抑制,Ki值为21.49μg/mL;乙酸乙酯及正丁醇萃取部分具有较强抗氧化作用,0.42μg/mL的乙酸乙酯及正丁醇萃取部分对DPPH.自由基的清除率分别可达到88.5%、93.6%,与0.3 mg/mL的抗坏血酸对DPPH.自由基的清除率96.3%相当;乙酸乙酯萃取部分对氧自由基亦有一定的清除作用,IC50为0.27 mg/mL。结论:西藏绿萝花醇提取物的乙酸乙酯萃取部分有较强的α-葡萄糖苷酶抑制及抗氧化作用;正丁醇萃取部分有较强的抗氧化作用。
OBJECTIVE: To study the inhibitory and anti-oxidative effects of extract of Ligustrum lucidum on α-glucosidase. Methods: 70% ethanol extract from Tibet green flower was extracted with petroleum ether, ethyl acetate and n-butanol, and the inhibitory and anti-oxidative effects of α-glucosidase in each part were determined. Results: The extracted fraction of ethyl acetate had a stronger inhibitory effect on the activity of α-glucosidase with IC50 of 43.63μg / mL and IC50 value of 250μg / mL lower than that of acarbose inhibiting α-glucosidase , The kinetics of inhibition showed competitive inhibition, Ki value was 21.49μg / mL; ethyl acetate and n-butanol extract had strong anti-oxidation effect, 0.42μg / mL of ethyl acetate and n-butanol extract part of DPPH .The free radical scavenging rate reached to 88.5% and 93.6% respectively, which was equivalent to 96.3% DPPH radical scavenging activity of 0.3 mg / mL ascorbic acid.Ethyl acetate extract also had certain scavenging effect on oxygen free radicals, IC50 is 0.27 mg / mL. CONCLUSION: The ethyl acetate fraction of the ethanolic extract of P. carota L. has a strong inhibitory and anti-oxidative effect on α-glucosidase. The n-butanol extraction has a strong anti-oxidant effect.
