目的:寻求提取高质量红豆杉毛状根基因组DNA的方法。方法:采用改进CTAB法提取发根农杆菌R1000诱导的红豆杉毛状根的基因组DNA,进行紫外分光检测、电泳分析及特异PCR扩增。结果:提取的DNA质量较好,纯度较高,可以满足进一步实验的要求。结论:运用改进CTAB法提取红豆杉毛状根基因组DNA,比传统方法更好地减少了基因组DNA中多糖、酚及次生代谢物的污染,不失为一种较好的提取方法。
Objective: To find a method for extracting high quality genomic DNA from hairy root of Taxus chinensis. Methods: The genomic DNA of hairy roots of Taxus chinensis was extracted by modified CTAB method and detected by UV spectrophotometry, electrophoresis and specific PCR. Results: The quality of extracted DNA was better and the purity was higher, which could meet the requirements of further experiments. Conclusion: It is a better method to extract genomic DNA from hairy roots of Taxus chinensis by improved CTAB method, which can reduce the pollution of polysaccharides, phenols and secondary metabolites in genomic DNA better than traditional methods.
