目的探讨P16、Caspase-3基因在葡萄胎发病机制中的作用。方法采用免疫组化二步法,对46例葡萄胎首次清宫标本(葡萄胎组)和20例早孕正常绒毛组织(正常组)中P16和Caspase-3进行检测。结果在早孕绒正常毛组织中Caspase-3表达全部阴性,而P16阳性率为80.00%。在葡萄胎组中P16、Caspase-3表达阳性率分别89.13%、60.87%。在非恶变葡萄胎与以后发生恶变的葡萄胎中两者表达均呈下降趋势。结论 P16、Caspase-3表达缺失可能在葡萄胎恶性转化中起重要作用。检测葡萄胎首次清宫标本P16及Caspase-3表达,可能在判断葡萄胎预后方面具有重要意义。
Objective To investigate the role of P16 and Caspase-3 in the pathogenesis of hydatidiform mole. Methods Immunohistochemical two-step method was used to detect the expression of P16 and Caspase-3 in 46 cases of first trimester of the mole (hydatidiform mole) and in 20 cases of normal early trimester of the primiparous tissue (normal group). Results The expression of Caspase-3 in normal plush normal hair tissue was all negative, while the positive rate of P16 was 80.00%. The positive rates of P16 and Caspase-3 in hydatidiform mole were 89.13% and 60.87% respectively. In non-malignant hydatidiform mole and subsequent hydatidiform mole in both expression showed a downward trend. Conclusion The loss of P16 and Caspase-3 expression may play an important role in the malignant transformation of hydatidiform mole. Detection of hydatidiform mole for the first time in patients with P16 and Caspase-3 expression, may be in determining the prognosis of hydatidiform mole is of great significance.
