目的改良大鼠双侧颈总动脉永久性阻断模型(2VO)以适应急性全脑缺血研究。方法平行制作双侧颈总、颈内、颈外动脉永久性阻断(6VO)和2VO全脑缺血模型及假手术组,比较2种模型的死亡率、行为学评分、海马CA1区细胞坏死率、凋亡率及炎症因子的差异。结果 6VO组24 h内死亡率(40%)与2VO组(0)、假手术组相比均明显增高,各个时间点行为学评分增高,神经元坏死、凋亡增加且炎症因子表达增高(均P<0.05)。结论改良模型可加重大鼠脑损伤,适合急性全脑缺血研究。
Objective To improve the rat bilateral common carotid artery occlusion model (2VO) to adapt to acute global cerebral ischemia. Methods The bilateral common carotid and internal carotid artery and internal carotid artery occlusion (6VO) and 2VO model rats were randomly divided into sham operation group and sham operation group. The mortality, behavioral score, hippocampal CA1 cell necrosis Rate, the rate of apoptosis and the difference of inflammatory factors. Results The intracerebral hemorrhage rate (40%) in 6VO group was significantly higher than that in 2VO group (0) and sham operation group at each time point, the behavioral score increased, neuronal necrosis, apoptosis increased and inflammatory factor expression increased P <0.05). Conclusion Improved model can increase brain injury in rats, suitable for acute global cerebral ischemia.
