目的:探讨不同浓度的当归贝母苦参丸含药血清抑制胃癌SGC-7901、MKN-45细胞增殖作用及其相关机制。方法:不同浓度含药血清处理SGC-7901细胞后,(1)MTT法测定细胞活力;(2)流式细胞术检测细胞周期;(3)Transwell侵袭实验及划痕实验观察侵袭能力和转移能力。结果:不同浓度的当归贝母苦参丸含药血清均能抑制细胞增殖,在10%浓度时抑制作用达最大值;侵袭及划痕实验示:含药血清干预后细胞侵袭及转移能力减弱;流式细胞术检测显示:含药血清干预后细胞S期比例下降,G0~G1期比例上升。结论:当归贝母苦参丸含药血清可以抑制SGC-7901细胞增殖,减弱其侵袭转移能力,其机制可能与其干预细胞周期有关。
OBJECTIVE: To investigate the effects of different concentrations of Radix Angelica Sinensis Seeds on the proliferation of gastric cancer SGC-7901 and MKN-45 cells and its related mechanisms. Methods: After treated with different concentrations of serum containing SGC-7901 cells, (1) MTT assay was used to determine cell viability; (2) flow cytometry was used to detect cell cycle; (3) Transwell invasion assay and scratch assay were used to observe the invasion and metastasis . Results: Different concentrations of Radix Angelicae Sinensis and Radix Cynanchi Lourin could inhibit the proliferation of cells, and reached the maximum value at 10% concentration. The invasion and scratching experiments showed that the cell invasion and metastasis abilities decreased after intervention with serum. The results of flow cytometry showed that the percentage of S phase decreased and the proportion of G0 to G1 phase increased after intervention with serum. CONCLUSION: Radix Angelica sinensis, Radix Salviae Miltiorrhizae, can inhibit the proliferation of SGC-7901 cells and decrease the invasion and metastasis of SGC-7901 cells. Its mechanism may be related to the intervention of cell cycle.
