为揭示土壤初始pH值、温度和有机质含量三因素对多粘类芽孢杆菌(Paenibacillus polymyxa)L_1-9菌株在黄瓜根际土壤定殖的影响,采用室内盆栽实验,在黄瓜一叶期灌根接种生防菌L_1-9发酵液,设置土壤pH值、温度和有机质含量的不同处理,培养并定期采集黄瓜根际土壤样品,采用实时荧光定量PCR技术检测海洋生防菌L_1-9在黄瓜根际土壤定殖的数量变化.定量数据及其显著性检验结果表明:温度、初始pH值和有机质含量显著影响L_1-9菌株在黄瓜根际土壤中的定殖.土壤初始pH为6、7和8均可定殖.初始pH值为7和8的处理,在多数调查时间内无显著差异,最大为每克干土(8.52±0.18)×10~9拷贝.在存在显著性差异的时间段内,初始pH值为6时,L_1-9菌株在黄瓜根际土壤中数量在同期中最大.15-30℃培养时,最大数量为每克干土(2.13±0.31)×10~(10)拷贝.温度越高,定殖越早,但在较高温度下定殖数量下降较快.高有机质含量可以增加定殖数量,提高定殖能力,最大数量为每克干土(1.15±0.04)×10~(10)拷贝.本研究表明实时荧光定量PCR技术适用于研究土壤根际生防菌的定殖;海洋生防菌L_1-9能够在较宽温度、初始pH值和有机质含量范围内稳定定殖,较低温度、中性偏酸初始pH值和高有机质含量更有利于L_1-9菌株在黄瓜根际土壤中定殖.图1表3参20
