目的:通过对鼻息肉组织中Fas和FasL因子的检测,以探讨Fas和FasL因子在鼻息肉发生,发展过程中的介导及调控作用。方法:应用逆转录聚合酶链反应(RTPCR)方法,检测40例鼻息肉组织匀浆及5例正常鼻黏膜中Fas和FasL因子的基因表达。结果:40例鼻息肉组织中FasL呈强势表达,Fas呈弱势表达,其相对系数的平均指数分别为:(0.62±0.74)、(0.14±0.25),与正常对照组相比差别有统计学意义(均P<0.05)。Fas在正常鼻腔黏膜上皮细胞中的表达阳性率显著大于鼻息肉组织,FasL不表达。结论在鼻息肉的发生、发展过程中存在着Fas的表达下调和FasL的上调,鼻息肉上皮细胞逃避了Fas和FasL系统介导的细胞凋亡而过度增殖。
OBJECTIVE: To detect the Fas and FasL factors in nasal polyps to investigate the role of Fas and FasL in the pathogenesis and progression of nasal polyps. Methods: The gene expressions of Fas and FasL in 40 cases of nasal polyps and 5 cases of normal nasal mucosa were detected by reverse transcriptase polymerase chain reaction (RTPCR). Results: FasL was strongly expressed in 40 cases of nasal polyps, while Fas was weakly expressed. The average relative coefficients of the two groups were (0.62 ± 0.74) and (0.14 ± 0.25) respectively, which were significantly different from those of the normal control group (All P <0.05). The positive expression rate of Fas in normal nasal mucosa epithelial cells was significantly higher than that in nasal polyps, but FasL was not expressed. Conclusions There is down-regulation of Fas expression and FasL up-regulation during the development of nasal polyps. Nasal polyp epithelial cells escape from excessive apoptosis by Fas and FasL-mediated apoptosis.
