为研究PML-RARα融合基因的反应核酸(ASODN)与维甲酸(ATRA)联合应用对NB4细胞诱导分化作用,探讨提高维甲酸疗效及逆转维甲酸耐药的方法。通过细胞生长曲线、形态学、细胞表面标志及NBT还原试验判定NB4细胞的生长、分化及功能,细胞周期运用流式细胞仪进行分析。结果显示,反义核酸与维甲酸联合应用对NB4细胞生长抑制和诱导分化作用增强,细胞周期中的S期细胞明显降低。证明了反义核酸与维甲酸具有协同作用,能够提高维甲酸的疗效。
In order to investigate the effect of the combination of AMLDN and ATRA on the differentiation of NB4 cells, the methods of improving the efficacy of retinoic acid and reversing the resistance of retinoic acid were studied. The growth, differentiation and function of NB4 cells were determined by cell growth curves, morphology, cell surface markers, and NBT reduction assays. The cell cycle was analyzed by flow cytometry. The results showed that the combination of antisense nucleic acid and retinoic acid enhanced the growth inhibition and differentiation of NB4 cells, and the S phase cells in the cell cycle decreased significantly. It has been demonstrated that antisense nucleic acids have a synergistic effect with retinoic acid and can increase the therapeutic effect of retinoic acid.
