目的 :探讨组织工程血管全生物支架的免疫原性的检测方法。 方法 :取猪颈总动脉 ,剥离外膜 ,剪成 2 cm长的血管段 ,随机分为 3组 :对照组、脱细胞组和脱细胞加碱处理组 ,每组 5段。以免疫组化方法检测血管脱细胞处理前后主要组织相容性复合体 (MHC )的变化情况 ,并将检测结果作图像分析。 结果 :对照组血管壁 MHC 高表达 ,脱细胞处理后 MHC 明显减少。图像分析得出阳性颗粒面积与视场面积比 :对照组为 (2 4 .0 9± 6 .0 9) % ,脱细胞组为 (6 .71± 2 .1 8) % ,脱细胞加碱处理组为 (1 .6 2± 0 .76 ) % ,组间比较差别均有显著意义 (P<0 .0 1 )。 结论 :MHC 免疫组化方法可检测血管生物支架抗原
Objective: To explore the method of detecting the immunogenicity of tissue engineering vascular bioscaffolds. Methods: The common carotid arteries were dissected and the adventitia were dissected. The vascular segments were cut into 2 cm long and randomly divided into 3 groups: control group, acellular group and acellular group. The changes of major histocompatibility complex (MHC) before and after vascular decellularization were detected by immunohistochemistry and the results were analyzed by image analysis. Results: The expression of MHC in the vascular wall of the control group was high, and the MHC decreased significantly after the decellularization. Image analysis showed that the ratio of the area of positive particles to the area of field of view was (24.0 ± 0.9)% in the control group and (6.71 ± 2.18)% in the acellular group, The treatment group was (1.62 ± 0.76)%, with significant difference between the two groups (P <0.01). Conclusion: MHC immunohistochemistry can detect the biologic scaffolds of blood vessels
