目的采用超高效液相色谱-四级杆串联飞行时间质谱法(UPLC-Q-TOF-MS)对亚香棒虫草提取物的主要化学成分进行分析鉴定。方法采用Agilent SB-C18(50 mm×4.6 mm,1.8μm)色谱柱,乙腈-水(0.1%甲酸)溶液为流动相梯度洗脱,流速0.25 m L/min,柱温30℃;信息关联模式(IDA)正、负离子分别采集。结果结合Peak View软件的Formula Finder等功能、数据库及二级碎片裂解规律,共鉴定出26个化合物,其中有9个为虫草中未曾报道物质,且含2个未知化合物;其主要化学成分包括生物碱、糖苷、核苷、氨基酸等成分。结论该方法精确、可靠、高效,适用于亚香棒虫草成分的快速鉴定,为亚相棒虫草的开发利用以及阐明其药效物质基础提供参考。
OBJECTIVE To analyze and identify the main chemical constituents of the extract of Cordyceps militaris by UPLC-Q-TOF-MS. Methods The mobile phase of acetonitrile-water (0.1% formic acid) was used as a mobile phase with an Agilent SB-C18 (50 mm × 4.6 mm, 1.8 μm) column with a flow rate of 0.25 m L / min and a column temperature of 30 ° C. (IDA) positive and negative ions were collected. Results Based on the functions of Peak Fin software such as Formula Finder, database and secondary fragmentation, a total of 26 compounds were identified. Among them, 9 were not reported in Cordyceps, and contained 2 unknown compounds. The main chemical components included biological Alkali, glycosides, nucleosides, amino acids and other ingredients. Conclusion The method is accurate, reliable and efficient and is suitable for the rapid identification of the constituents of Cordyceps militaris. It provides a reference for the development and utilization of Cordyceps subtilis and elucidates its pharmacological basis.
