目的 :探讨胃癌及淋巴结转移癌组织中p28、基质金属蛋白酶2(matrix metalloproteinase 2,MMP2)和β-catenin蛋白表达的差异,并结合肿瘤的病理学特征进行分析。方法:收集123例胃腺癌组织(包括41例不伴淋巴结转移的胃癌组织,82例伴淋巴结转移的胃癌组织及相应的淋巴结转移癌组织)构建组织芯片,采用免疫组织化学方法检测其p28、MMP2和β-catenin蛋白的表达情况。结果:1伴淋巴结转移的胃癌组织中的p28和MMP2阳性表达均明显高于无淋巴结转移的胃癌组织(P<0.05)。2伴淋巴结转移的胃癌病例中,p28与MMP2、p28与β-catenin在原发癌组织及淋巴结转移癌组织中的共表达均呈明显正相关(P<0.05)。3β-catenin在伴淋巴结转移和无淋巴结转移胃癌组织中的阳性表达差异尚无统计学意义(P>0.05)。结论:p28和MMP2高表达的胃癌易发生淋巴结转移;p28与MMP2、p28与β-catenin都具协同性,可能共同参与了胃癌的发生和发展。
Objective: To investigate the differences of p28, matrix metalloproteinase 2 (MMP2) and β-catenin protein expression in gastric cancer and lymph node metastasis and to analyze their pathological characteristics. Methods: Tissue microarrays were collected from 123 gastric adenocarcinoma tissues (including 41 gastric cancer without lymph node metastasis, 82 gastric cancer with lymph node metastasis and corresponding lymph node metastasis). The expression of p28, MMP2 And β-catenin protein expression. Results: 1 The positive expression of p28 and MMP2 in gastric cancer with lymph node metastasis were significantly higher than those without lymph node metastasis (P <0.05). 2 cases of gastric cancer with lymph node metastasis, p28 and MMP2, p28 and β-catenin in primary cancer and lymph node metastasis co-expression were positively correlated (P <0.05). The positive expression of 3β-catenin in gastric cancer with lymph node metastasis and non-lymph node metastasis was not statistically different (P> 0.05). CONCLUSIONS: Lymphatic metastasis is easily observed in gastric cancer with high expression of p28 and MMP2. Both p28, MMP2, p28 and β-catenin are synergistic and may participate in the occurrence and development of gastric cancer.
