目的:检测miR-21、miR-192和miR-200c 3种微小RNA(miRNA)在糖尿病肾病(DN)患者血清中的含量,评价其在DN诊断中的价值。方法:选择2型DN患者36例,正常对照组26例。收集临床血清样本,应用荧光定量PCR方法检测相应miRNA的含量,应用ROC曲线评价三者诊断DN的敏感度。结果:①糖尿病患者血清中miR-21和miR-192的水平均较对照组显著升高(P均<0.05),而miR-200c较对照组显著下降(P<0.05)。②miR-21、miR-192和miR-200c ROC曲线下的面积分别是0.822、0.736和0.751。结论:糖尿病患者血清中,miR-21,miR-192和miR-200c是早期诊断DN的敏感指标,有望用于临床监测。
Objective: To detect the levels of miR-21, miR-192 and miR-200c in patients with diabetic nephropathy (DN) and evaluate their value in the diagnosis of DN. Methods: 36 cases of type 2 DN patients and 26 cases of normal control group were selected. Clinical serum samples were collected, the corresponding miRNA content was detected by real-time PCR, and the sensitivity of the three methods for diagnosis of DN was evaluated by ROC curve. Results: ① The serum levels of miR-21 and miR-192 in diabetic patients were significantly higher than those in control group (all P <0.05), while miR-200c was significantly lower than that in control group (P <0.05). The area under ROC curve of miR-21, miR-192 and miR-200c were 0.822, 0.736 and 0.751, respectively. Conclusion: The serum of patients with diabetes, miR-21, miR-192 and miR-200c is a sensitive indicator of early diagnosis of DN, is expected to be used for clinical monitoring.
