利用31P NMR谱对同一样品中ATP、ADP、AMP以及磷酸钠和焦磷酸钠等含磷物质进行定性定量分析,得到ATP、ADP、AMP在0.004~0.080 mol·L-1,Na H2PO4、Na4P2O7在0.005~0.100 mol·L-1内具有很好的线性关系。该方法精密度为0.40%~1.30%,添加回收率为96.9%~105.2%。把该方法应用于ATP注射液的测定,测得ATP注射液中的杂质为ADP和磷酸盐,ATP的含量符合药典要求。实验结果表明该方法重现性好、准确度高,而且无需前处理、操作简单、快速、无其他元素的干扰,是一种全新、可靠的分析ATP、ADP、AMP以及磷酸盐的方法。
The content of ATP, ADP and AMP in 0.004-0.080 mol·L-1 ATP, ADP, AMP and other phosphorus-containing substances such as sodium phosphate and sodium pyrophosphate in the same sample were determined by 31P NMR spectroscopy. 0.005 ~ 0.100 mol · L-1 has a good linear relationship. The precision of the method was 0.40% ~ 1.30%, and the recovery rate was 96.9% ~ 105.2%. The method was applied to the determination of ATP injection. The impurities in the ATP injection were measured as ADP and phosphate, and the content of ATP was in accordance with the requirements of Pharmacopoeia. The experimental results show that this method is a new and reliable method for the analysis of ATP, ADP, AMP and phosphate because of its good reproducibility, high accuracy, no pretreatment, simple operation, fast operation and no interference with other elements.
