目的 :研究缺血再灌注损伤后肠粘膜上皮细胞凋亡与 ICE和 bcl 2基因 m RNA表达的关系 ,并探讨药用大黄的肠道屏障保护作用机制。方法 :制备小鼠肠系膜上动脉 (SMA)缺血再灌注损伤的实验模型 ,琼脂糖凝胶电泳法检测肠上皮细胞凋亡 ,逆转录定量多聚酶链式反应 (RT Q PCR)检测肠上皮细胞中 ICE和bcl 2 m RNA的表达。结果 :大黄治疗组小鼠肠上皮细胞致伤后各时间点 DNA梯状电泳条带强度显著弱于对照组小鼠。 bcl 2基因在 2组小鼠肠上皮细胞各时间点均呈低水平表达 ;而大黄治疗组 ICE基因的表达与对照组相比均有显著性下降 (P均 <0 .0 5 )。结论 :缺血再灌注损伤过程中 ,ICE基因的持续高水平表达可能是导致肠上皮细胞发生异常凋亡的重要原因 ,致伤后立即应用大黄煎剂可以下调肠上皮细胞 ICE基因的 m RNA表达 ,减轻肠上皮细胞的凋亡程度。
Objective: To study the relationship between apoptosis of intestinal epithelial cells and expression of ICE and bcl 2 gene m RNA after ischemia-reperfusion injury, and to explore the protective mechanism of intestinal barrier in medicinal rhubarb. METHODS: An experimental model of mouse superior mesenteric artery (SMA) ischemia-reperfusion injury was prepared. Agarose gel electrophoresis was used to detect intestinal epithelial cell apoptosis. Reverse transcription quantitative polymerase chain reaction (RT Q PCR) was used to detect intestinal epithelial cells. Expression of ICE and bcl 2 m RNA. Results : The intensity of DNA ladder electrophoresis was significantly weaker than that of control mice at various time points after the injury of intestinal epithelial cells in mice treated with rhubarb. The expression of bcl 2 gene in intestinal epithelial cells of the two groups was low at all time points, but the expression of ICE gene in the rhubarb treatment group was significantly lower than that of the control group (P<0.05). Conclusion: The persistent high-level expression of ICE gene may be an important cause of abnormal apoptosis in intestinal epithelial cells during ischemia-reperfusion injury. Rhubarb decoction can reduce the m RNA expression of ICE gene in intestinal epithelial cells immediately after injury. , to reduce the degree of apoptosis of intestinal epithelial cells.
