目的:应用响应曲面分析法观察归芎药对不同制法、不同配比对DPPH自由基清除率体外抗氧化活性的相互作用研究。方法:采用2,2-二苯基-1-苦肼基(DPPH)自由基清除率法测定当归-川芎10个配比,3种不同提取方法的提取物的抗氧化活性,应用非线性回归的方法确定量效曲线的各个参数,建立三维响应面模型,应用Matlab软件构建响应曲面的三维图形。结果:不同配比不同制法的30个样品,均具有明显的清除自由基活性,且表现出明显的量效关系。水提液的协同作用范围窄,几乎成点状分布,拮抗作用范围呈6条宽窄不一的条带状分布;醇提液的协同作用呈小条状分布,拮抗作用呈点状分布。在先水提后醇提液协同作用呈条状分布(3条),拮抗作用呈点状分布。总之先水提后醇提液的协同作用区域大于醇提液,水提液次之;3种提取方法大部分区域均未表现出明显协同与拮抗作用。结论:采用响应曲面分析法定量研究中药配伍药物相互作用,为相关研究提供了思路与方法的参考。
OBJECTIVE: To observe the effect of ZXX on the anti-oxidative activity of DPPH free radical scavenging in vitro by different methods and different proportions by response surface methodology. Methods: The anti-oxidative activities of extracts from Angelica sinensis-Ligusticum chuanxiong 10 and three different extraction methods were determined by 2,2-diphenyl-1-picrylhydrazyl radical scavenging assay (DPPH) Method to determine the parameters of the dose-response curve, the establishment of three-dimensional response surface model, the application of Matlab software to build three-dimensional response surface graphics. Results: All 30 samples with different formulation methods had obvious scavenging free radical activity and showed a significant dose-response relationship. The synergistic effect range of aqueous extract was narrow, almost in a dotted distribution, and the range of antagonistic effect was six bands with different widths and widths. The synergistic effect of alcohol extracts showed a strip-like distribution and the antagonistic effect showed a dot-like distribution. In the previous study, the synergistic effect of ethanol extract showed a strip-like distribution (3 strips), and the antagonistic effect showed a dot-like distribution. In conclusion, the synergistic effect of alcohol extraction was greater than that of alcohol extraction and water extraction followed by water extraction. No significant synergy and antagonism were observed in most of the three extraction methods. Conclusion: Using response surface methodology to quantitatively study the compatibility of traditional Chinese medicine (TCM) drugs provides a reference for the related research.
