目的 对人睾丸 L ipocalin型前列腺素 D合成酶 (L - PGDS) c DNA进行克隆和序列分析。 方法 用人脑 L - PGDS的特异性引物对人睾丸 RNA进行逆转录和 PCR扩增 ,并对扩增产物进行纯化和测序。 结果 人睾丸 L - PGDS c DNA核苷酸序列与人外周血淋巴细胞的 L - PGDS基因编码区序列的一致性为 99.5 % ,与人脑 L -PGDS c DNA的一致性为 96 .0 % ,相应氨基酸的一致性分别为 10 0 %和 94.2 %。 结论 人睾丸 L - PGDS c DNA序列及推导的氨基酸序列的阐明 ,对进一步探讨 L - PGDS蛋白的特性及其在男性生殖中的作用是十分有意义的。
Objective To clone and sequence the c - DNA of human testis Lipocalin - type prostaglandin D synthase (L - PGDS). Methods The human testicular RNA was reverse transcribed and amplified by PCR using human L - PGDS specific primers. The amplified product was purified and sequenced. Results The nucleotide sequence of L - PGDSc DNA in human testis was 99.5% identical to that of L - PGDS gene in human peripheral blood lymphocytes, and 96.0% in L - PGDS DNA. The corresponding amino acid identities were 100% and 94.2%, respectively. Conclusion The elucidation of L - PGDSc DNA sequence and deduced amino acid sequence of human testis is of great significance to further explore the characteristics of L - PGDS protein and its role in male reproduction.
