目的研究羟基红花黄色素A(HSYA)联用β-乳香酸(β-BA)对血瘀证模型大鼠凝血功能和全血粘度的影响。方法将40只SD大鼠随机分为空白对照组(等体积生理盐水)、模型组(等体积生理盐水)、HSYA(100 mg/kg)组、β-BA(200 mg/kg)组、联合用药(HSYA 50 mg/kg+β-BA 100 mg/kg)组,每12h给药1次,连续7 d。第5次给药后给予大鼠皮下注射盐酸肾上腺素(0.7 mg/kg)2次,间隔4 h,中间给予冰水刺激以复制大鼠急性血瘀证模型。末次给药30 min内取腹主动脉血检测凝血指标和不同切变率下的全血粘度指标(WBV)。结果与空白对照组比较,模型组大鼠1 s,5 s,30 s,200 s切变率全血粘度普遍升高,血浆凝血酶时间(TT)、血浆凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)缩短,纤维蛋白原(FIB)含量增加,差异均有统计学意义(P<0.05)。与模型组比较,HSYA组、β-BA组、联合用药组大鼠1s,5s,30s,200s切变率全血粘度降低,血浆TT、PT、APTT延长,FIB含量减少,差异均有统计学意义(P<0.05)。结论 HSYA联用β-BA能显著改善血瘀证大鼠凝血功能和在不同切变率下的全血粘度。
Objective To investigate the effect of hydroxysafflor yellow A (HSYA) combined with β-boswellic acid (β-BA) on the coagulation function and whole blood viscosity in blood stasis model rats. Methods 40 SD rats were randomly divided into blank control group (equal volume of normal saline), model group (equal volume of normal saline), HSYA group (100 mg / kg) and β-BA (200 mg / Administration (HSYA 50 mg / kg + β-BA 100 mg / kg) was given once every 12 hours for 7 days. After the fifth administration, rats were subcutaneously injected with epinephrine hydrochloride (0.7 mg / kg) 2 times at intervals of 4 h, and ice-water stimulation was given in the middle to replicate the model of acute blood stasis in rats. Abdominal aorta blood was taken within 30 min after the last administration to measure blood coagulation index and whole blood viscosity index (WBV) at different shear rates. Results Compared with the blank control group, the whole blood viscosity of the rats in the model group at 1, 5, 30, and 200 s were generally increased. The plasma thrombin time (TT), plasma prothrombin time (PT) and activation Partial thromboplastin time (APTT) shortened, fibrinogen (FIB) increased, the difference was statistically significant (P <0.05). Compared with the model group, the viscosity of whole blood in 1s, 5s, 30s, 200s of HSYA group, β-BA group and combined treatment group decreased, the plasma TT, PT, APTT prolonged and the FIB content decreased Significance (P <0.05). Conclusion HSYA combined with β-BA can significantly improve the blood coagulation function and blood viscosity in different shear rate of whole blood viscosity.
