在法国Bordeaux大学和France’s INRA,M.Brison及其同事发现可用冷冻保存法消除实验污染的李属植物Prunus根状茎中的病毒。Brison等人特意用洋李痘病毒(Plum Pox Poty)感染离体生长的洋李根状茎。被污染植物的茎尖在加入了5%二甲亚砜和2%脯氨酸的预培养基中于4℃培养24小时,然后用经修饰的PVS2低温防护溶液处理并在浸入液氮前按1℃/mm冷却至-40℃。对照茎尖在分生组织培养基上培养不经冷冻。4个月后,检测茎尖中洋李痘病毒是否存在。作者报道,在一般情况下,20%的对照茎尖不存在病毒污染。小茎尖(0.3~0.5mm)的结果最好。相反,50%的冷冻保存植物没有病毒而且这一比例不受茎尖的大小影响。 Brison等人总结出:对于种质保存这是又一有用的方法,与单纯的分生组织培养相比冷冻
At Bordeaux University in France and France’s INRA, M. Brison and colleagues found that cryopreservation can be used to eliminate the experimentally contaminated Prunus rhizome virus from Prunus.
