目的:优选超临界CO2流体萃取法萃取丹葶肺心颗粒中挥发油的工艺条件。方法:以萃取压力、萃取温度、萃取时间3个因素为主要影响因素,以鱼腥草素得率、鱼腥草挥发油得率、枳壳挥发油得率、总评归一值(OD)为评价指标进行Box-Behnken设计试验,考察最佳萃取工艺。结果:优化的最佳工艺条件为:萃取压力27.5 MPa,萃取温度58℃,萃取时间105 min,此时鱼腥草素、鱼腥草挥发油、枳壳挥发油得率分别为0.061%、1.973%,0.752%;对最佳工艺进行验证,结果实测平均值与模型预测值非常接近,表明该模型较可靠。结论:采用Box-Behnken设计方法科学、合理、可行;超临界萃取效率高,稳定性好,在中药复方挥发油的提取方面有相当大的发展潜力。
Objective: To optimize the extraction of volatile oil from Danjianxiongxin granules by supercritical CO2 fluid extraction. Methods: The factors of extraction pressure, extraction temperature and extraction time were taken as the main factors. The yield of Houttuynia cordata, the yield of volatile oil of Houttuynia cordata, the yield of volatile oil of Fructus Aurantii, Box-Behnken design test to investigate the best extraction process. Results: The optimal extraction conditions were extraction pressure of 27.5 MPa, extraction temperature of 58 ℃ and extraction time of 105 min. The volatile oil yield of Houttuynia cordata, Houttuynia cordata and Fructus Aurantii were 0.061% and 1.973%, respectively. 0.752%. The best process was verified. The results showed that the average of the measured values was very close to the predicted value of the model, indicating that the model is more reliable. Conclusion: The Box-Behnken design method is scientific, reasonable and feasible. The supercritical extraction has high efficiency and good stability. It has considerable potential in the extraction of volatile oil from traditional Chinese medicine.
