目的:观察金翘热毒清颗粒和金翘热毒清喷雾干燥粉体内、外的抗流感病毒作用,并比较不同干燥工艺制备的金翘热毒清抗病毒作用差异。方法:体外采用四氮唑蓝(MTT)比色法测定样品以不同给药方式(药物直接作用于病毒、药物直接作用于病毒感染的细胞)抑制甲型流感病毒鼠肺适应株(FM1)的半数抑制浓度(IC50),并计算治疗指数(TI)。狗肾细胞密度为1×104/孔,金翘热毒清喷雾干燥粉质量浓度为(2.61,1.31,0.66,0.16 g·L-1),金翘热毒清颗粒(6.01,3.05,1.52,0.76 g·L-1),作用时间为5 d,每组4个样本。将小鼠随机分为对照组,利巴韦林组(0.07 g·kg-1),金翘热毒清颗粒(按生药量计)组(11.7 g·kg-1)、金翘热毒清喷雾干燥粉(按生药量计)组(5.85,11.7,23.4 g·kg-1),采用流感病毒株FM1滴鼻感染小鼠建立感染模型,感染2 h后连续ig给药5 d,每天2次,以生命延长率和肺指数抑制率为指标,评价二者的保护作用。结果:在药物直接作用于病毒组和药物直接作用于病毒感染的细胞组中,金翘热毒清喷雾干燥粉的IC50分别为0.77,0.80 g·L-1,TI分别为11.78,11.34;金翘热毒清颗粒的IC50分别为3.70,3.60 g·L-1,TI分别为6.37,6.55;金翘热毒清喷雾干燥粉23.4 g·kg-1,其生命延长率为60.81%,肺指数抑制率为13.18%,与病毒组比较,差异均有统计学意义(P<0.05);金翘热毒清颗粒与金翘热毒清喷雾干燥粉相同剂量下的实验结果平行比较,未见显著差异。结论:金翘热毒清颗粒与金翘热毒清喷雾干燥粉都有抗流感病毒作用;体外在相同剂量下金翘热毒清喷雾干燥粉对流感病毒株的抑制作用优于金翘热毒清颗粒。
Objective: To observe the anti-influenza virus effect of Jinqiao Reutuan Granule and Jinqiao Renduanqing spray-dried powder, and to compare the anti-virus effect of Jinqiao Reituqing prepared by different drying processes. Methods: MTT assay was used to determine the inhibitory effect of influenza virus type 1 (FM1) in mice by different modes of administration (the drugs act directly on the virus and the drugs act directly on the virus-infected cells) Half inhibitory concentration (IC50) and calculate the therapeutic index (TI). The mice were infected with influenza virus strain FM1 to infect the mice. Results: IC50 of Jinqiao Rendoxicate Spray Dried Powder was 0.77,0.80 g · L-1, TI was 11.78 and 11.34, respectively, when the drug directly acted on the virus group and the drug directly acted on the virus-infected cell group. The IC50 of QWD were 3.70 and 3.60 g · L-1, TI were 6.37 and 6.55, respectively.
