目的探讨阿仑膦酸钠(ALN)对骨髓间充质干细胞(BMSCs)增殖和骨向分化能力的影响,观察其促BMSCs骨向分化过程中对转化生长因子β1(TGF-β1)及骨形成蛋白-2(BMP-2)表达的影响,分析可能的作用途径。方法以碱性磷酸酶(ALP)等指标确定干预药物不同浓度梯度中最佳促BMSCs骨向分化剂量;分组实验,通过检测诱导分化过程中ALP和钙化结节等指标的表达,观察其对BMSCs骨向分化能力的影响;ELISA法检测骨向分化过程中TGF-β1、BMP-2的表达。结果 ALN最佳促BMSCs增殖和骨向分化浓度为1×10-8mol/L,该浓度ALN能促进成骨指标的表达和TGF-β1和BMP-2分泌增加;具有促BMSCs骨向分化能力的ALN各诱导组均伴随TGF-β1和BMP-2分泌增加。结论 ALN能促进BMSCs增殖和骨向分化;在BMSCs骨向分化过程中,上调TGF-β1、BMP-2的表达量可能是各干预药物促进MSCs骨向分化的作用机制之一。
Objective To investigate the effects of alendronate sodium (ALN) on the proliferation and differentiation of bone marrow mesenchymal stem cells (BMSCs), and to investigate the effect of transforming growth factor β1 (TGF-β1) and bone formation Protein-2 (BMP-2) expression, and analyze the possible ways of action. Methods Bone marrow differentiation dose of the best promoting BMSCs in different concentration gradient was determined by alkaline phosphatase (ALP) and other indicators. By grouping experiments, the expression of ALP and calcified nodules during induction differentiation was observed, Bone differentiation ability; ELISA method to detect bone differentiation into TGF-β1, BMP-2 expression. Results The proliferation and osteogenic differentiation of the best BMSCs induced by ALN was 1 × 10-8 mol / L, and the concentration of ALN could promote the expression of osteoblast and the secretion of TGF-β1 and BMP-2. ALN induced groups were accompanied by increased secretion of TGF-β1 and BMP-2. CONCLUSIONS: ALN can promote the proliferation and differentiation of BMSCs. Upregulation of the expression of TGF-β1 and BMP-2 during BMSCs differentiation may be one of the mechanisms by which each intervention can promote bone differentiation of MSCs.
