目的研究动力相关蛋白1(Drp1)在羊瘙痒因子139A感染的小鼠脑组织中的变化。方法采用Western Blot方法检测Drp1在羊瘙痒因子139A感染的脑组织匀浆中的含量变化及其亚硝基化水平。采用组织免疫荧光方法检测Drp1在脑组织中的分布。结果在羊瘙痒因子139A感染终末期小鼠脑组织匀浆中,Drp1总量略有降低。对感染不同时间点的动态分析结果显示Drp1含量呈逐渐降低趋势,终末期稍有回升。感染终末期脑组织中亚硝基化水平明显增高。组织免疫荧光显示正常和羊瘙痒因子139A感染终末期小鼠脑组织中,Drp1与神经元细胞存在共定位现象。结论Drp1在小鼠中枢神经系统中主要分布于神经元细胞,在羊瘙痒因子139A感染的小鼠脑组织中,Drp1总量略有降低,另外亚硝基化水平明显增高,提示在羊瘙痒病感染的小鼠脑组织中,线粒体动力相关蛋白的表达量及翻译后修饰水平出现了变化,在感染过程中起着重要作用。
Objective To investigate the changes of the brain-related protein 1 (Drp1) in mice infected with pruritus factor 139A. Methods Western Blot method was used to detect the changes of content of Drp1 in the homogenate of pruritus 139A-infected brain tissue and its nitrosylation level. Tissue immunofluorescence was used to detect the distribution of Drp1 in brain tissue. Results The total amount of Drp1 slightly decreased in the brain tissue homogenate of sheep with ITC-139A infection. Dynamic analysis of infection at different time points showed that the content of Drp1 gradually decreased, the end of a slight rebound. End-stage brain tissue infection nitrosylation levels were significantly higher. Tissue immunofluorescence showed normal co-localization of Drp1 with neuronal cells in normal and scrapie-infected 139A-infected mice. Conclusion Drp1 is mainly distributed in neuronal cells in the central nervous system of mice. The total amount of Drp1 in brain tissue of mice with pruritus 139A is slightly decreased, and the nitrosylation level is significantly increased, which indicates that in the scrapie In infected mouse brain tissue, the expression of mitochondria-related protein and the level of post-translational modifications have changed, which play an important role in the process of infection.
