背景:研究指出核因子?B在人根尖周炎组织中m RNA的表达水平明显高于正常根尖周组织。目的:观察核因子?B在大鼠实验性根尖周炎部位的表达。方法:取6周龄雄性SD大鼠20只,右侧下颌第1磨牙建立根尖周炎模型为实验组;左侧下颌第1磨牙为正常对照组。分别于术后1,2,3,4周随机取5只鼠,取下颌骨组织块制作冰冻切片。苏木精-伊红染色观察根尖区组织炎症的病理特征,免疫组织化学染色检测核因子?B在大鼠根尖周炎组织中的表达与分布。结果与结论:(1)苏木精-伊红染色见实验侧下颌第1磨牙根尖周组织炎性浸润,向周围牙槽骨扩散蔓延;正常对照组未见病理表现;说明建模成功;(2)免疫组织化学染色显示,造模后1,2,3,4周,实验组的根尖周炎症区域中均可见核因子?B呈阳性表达。从1周至2周其表达量增加,2周时核因子?B的表达量呈现高峰,3周开始降低,4周时最低,各个时间点的表达差异均有显著性意义(P<0.05);实验组的阳性细胞数均高于正常对照组(P<0.05);(3)结果表明,核因子?B的表达情况与根尖周炎的进程密切相关,提示核因子?B参与了根尖周炎的炎症反应过程。
BACKGROUND: The study indicates that the expression level of m RNA in nuclear pericytes is significantly higher than that in normal apical tissues. Objective: To observe the expression of nuclear factor-B in the experimental periapical periodontitis in rats. Methods: Twenty rats of 6-week-old male Sprague-Dawley rats and the right mandibular first molar were used to establish the model of periapical periodontitis. The left mandibular first molar was used as normal control group. Five rats were randomly selected at 1, 2, 3 and 4 weeks postoperatively, and the jaw tissue was removed to make frozen sections. The hematoxylin-eosin staining was used to observe the pathological features of tissue inflammation in the apical area. The expression and distribution of nuclear factor-B in the periapical periodontitis tissues were detected by immunohistochemical staining. RESULTS AND CONCLUSION: (1) Hematoxylin-eosin staining showed inflammatory infiltration in periapical tissues of the mandibular first molar on the experimental side and spread to the surrounding alveolar bone. No pathological findings were found in the normal control group, indicating successful modeling. (2) Immunohistochemical staining showed that at 1, 2, 3 and 4 weeks after model establishment, nuclear factor B was positive in the periapical inflammatory area of the experimental group. The expression of NF-κB increased from 1 week to 2 weeks. The expression of NF-κB peaked at 3 weeks, decreased at 3 weeks and lowest at 4 weeks (all P <0.05). (3) The results showed that the expression of nuclear factor-B was closely related to the process of apical periodontitis, suggesting that nuclear factor-B involved in the apical Inflammation of the inflammation process.
