目的:了解冬凌草甲素(Orid)及其它香茶菜属二萜类化合物香茶菜醛(Ame)毛叶香茶菜醇(Iso)是否与冬凌草甲素同样具有抗氧化作用。方法:用分光光度法测定脂质过氧化物丙二醛(MDA)含量及线粒体肿胀度。用荧光分光光度法测定线粒体膜流动性。结果:Orid,Ame,Iso40,80,160μmol/L抑制铁半胱氨酸(Fe2+Cys)引起的肝线粒体MDA形成,并呈剂量-依赖关系。Orid160μmol/L抑制肝线粒体膜流动性下降。(P值分别为0297±0.022,Fe2+Cys的P值为0423±0.014);Ame160μmol/L还可抑制脂质过氧化引起的肝线粒体肿胀。结论:Ame,Iso与Orid同样可能通过抑制脂质过氧化而产生抗氧化作用。
OBJECTIVE: To investigate whether Orid and other Chaenomeles diterpenoids, Ame, are the same antioxidants as oridonin. Methods: The contents of malondialdehyde (MDA) and mitochondrial swelling were measured by spectrophotometry. Mitochondrial membrane fluidity was determined by fluorescence spectrophotometry. RESULTS: Orid, Ame, Iso40, 80 and 160μmol / L inhibited the formation of hepatic mitochondrial MDA induced by iron-cysteine (Fe2 + -Cys) in a dose-dependent manner. Orid160μmol / L inhibited the decrease of mitochondrial membrane fluidity in the liver. (P = 0297 ± 0.022, P = 02323 ± 0.014, respectively). Ame160μmol / L also inhibited hepatic mitochondria swelling induced by lipid peroxidation. Conclusion: Ame, Iso and Orid may also have antioxidant effects by inhibiting lipid peroxidation.
