目的:促性腺激素释放激素拮抗剂(GnRHant)能降低小鼠糖尿病的发生率,文中在细胞水平研究GnRHant对In-R1-G9细胞内胰高血糖素(glucagon)表达的调控作用。方法:分别用终浓度为0.1、10、1 000 nmol/L的GnRHant处理培养的In-R1-G9细胞2 h(对照组用甘露醇处理),通过Western blot和荧光定量PCR技术观察胰高血糖素在蛋白水平及转录水平的变化。结果:与对照组相比,低浓度的GnRHant(0.1 nmol/L)使胰高血糖素在蛋白水平和转录水平的表达均有所降低,但是无统计学意义(P>0.05);而高浓度的GnRHant(10 nmol/L、1 000 nmol/L)能明显地降低胰高血糖素的表达(P<0.05)。结论:一定浓度的GnRHant能降低In-R1-G9细胞内胰高血糖素的表达。
OBJECTIVE: GnRHant can reduce the incidence of diabetes mellitus in mice. The regulation of GnRHant on the expression of glucagon in In-R1-G9 cells was studied at the cellular level. Methods: The cultured In-R1-G9 cells were treated with GnRHant at final concentration of 0.1, 10 and 1 000 nmol / L for 2 h, respectively. The control group was treated with mannitol.
