目的探究细胞培养液中糖浓度对人牙周膜成纤维细胞原代培养成功率的影响。方法取60颗健康牙,随机分成3组,每组20颗,分别采用低糖、高糖、低糖+甘露醇培养液对其牙周膜成纤维细胞进行原代培养。记录每组细胞原代培养成功数,以及首次传代时间、传代后获得稳定的成纤维细胞系的成功数。结果人牙周膜成纤维细胞原代培养的成功率依次为低糖:90.00%,高糖:60.00%,低糖+甘露醇:85.00%。结论低糖较高糖培养液更有利于提高人牙周膜成纤维细胞原代培养的成功率。
Objective To investigate the effect of glucose concentration in cell culture medium on the success rate of primary culture of human periodontal ligament fibroblasts. Methods Totally 60 healthy teeth were randomly divided into 3 groups with 20 in each group. Primary cultured periodontal ligament fibroblasts were cultured with low glucose, high glucose, low glucose and mannitol respectively. The number of successful primary culture of each group of cells was recorded, as well as the passage time of the first passage, and the number of successful fibroblast cell lines obtained after passage. Results The success rate of primary culture of human periodontal ligament fibroblasts was low glucose: 90.00%, high glucose: 60.00%, low glucose + mannitol: 85.00%. Conclusion Low glucose and high glucose culture medium is more beneficial to improve the success rate of human periodontal ligament fibroblast primary culture.
