目的:采用3种方法对五味子进行蛋白质提取,寻找一种简单有效的五味子蛋白提取方法。方法:利用SDS-聚丙烯酰胺凝胶电泳法对蛋白提取物进行分析,同时依据Lowry法对五味子总蛋白含量进行测定,HPLC测定五味子蛋白提取物中氨基酸的含量。结果:3种方法提取总蛋白质量分数分别为58.44%,19.21%,26.18%,其所含氨基酸质量分数分别为57.46%,15.41%,23.14%。结论:采用脱脂硫酸铵沉降法提取所得总蛋白及其所含氨基酸含量较高、纯度较合理,方法较可行。
OBJECTIVE: To extract the protein from Schisandra sphenanthera by three methods and to find a simple and effective method for extracting Schisandra protein. Methods: The protein extracts were analyzed by SDS-polyacrylamide gel electrophoresis, and the total protein content of Schisandra sphenanthera was determined by Lowry method. The content of amino acids in the extract of Schisandra chinensis was determined by HPLC. Results: The total protein contents of the three methods were 58.44%, 19.21% and 26.18%, respectively. The contents of amino acids in the three methods were 57.46%, 15.41% and 23.14%, respectively. Conclusion: The total protein extracted by ammonium sulfate desalting method and its content of amino acids contained in a higher purity and reasonable, the method is more feasible.
