目的 研究GNB3基因C82 5T多态性和硝苯地平控释片降压效果的相关性。方法 收集了 5 5 4个原发性高血压患者给予硝苯地平控释片治疗 ,通过问卷调查和随访取得其流行病学的临床资料 ,用PCR RFLP法测定其GNB3基因的基因型。用线性回归和Logistic回归分析了该基因型对疗效的影响。结果 82 5T等位基因在我们研究的高血压群体中的频率为 4 8 4 %。稍高于Siffert等报道的正常血压的中国人的T等位子频率 (42 % )。T等位子的携带者在硝苯地平控释片治疗 15d后 ,其收缩压下降幅度较CC纯合子小3 3mmHg。经多元线性回归分析两组间差别有显著性 (P<0 0 5 )。取收缩压下降值的分布曲线的上下 2 0 %分别作为疗效好与差组 ,经多元Logistic回归分析表明T等位子的携带者比CC基因型人群取得较好的收缩压治疗效果的可能性要小 ,OR值为 0 33,P =0 0 2。结论 GNB3基因C82 5T多态性和硝苯地平控释片的降压疗效相关
Objective To study the relationship between the C82 5T polymorphism of GNB3 gene and the antihypertensive effect of nifedipine controlled release tablets. Methods A total of 554 essential hypertension patients were treated with nifedipine controlled release tablets. The epidemiological data were obtained by questionnaire survey and follow-up. The genotypes of GNB3 gene were determined by PCR RFLP. The linear regression and Logistic regression analysis of the genotype on the effect. Results The frequency of the 5T allele in the hypertensive population we studied was 444%. Slightly higher than the T-allele frequency (42%) in Chinese subjects with normotensive reported by Siffert et al. T carriers such as seat in nifedipine controlled release tablets 15d after treatment, the systolic blood pressure decreased more than CC homozygotes small 3 3mmHg. Multivariate linear regression analysis showed significant difference between the two groups (P <0 05). Take the systolic blood pressure drop value of the distribution curve above and below 20% as a good effect and poor group, multivariate Logistic regression analysis showed that T-bit carriers of CC genotype better than the possibility of systolic blood pressure treatment effect Small, OR value is 0 33, P = 0 0 2.
