通过根癌农杆菌( Agrobacterium tumefaciens) 介导, 利用特异启动子PSAG12 与异戊烯基转移酶(Isopentenyltransferase,ipt) 基因构建的嵌合基因PSAG12ipt 对5 个番茄品种进行遗传转化, 结果共获得来自31 个不同外植体的44 株转化再生植株。感染子叶的分化再生及转化植株田间生长正常。对其中部分转化再生植株进行PCR 检测证明为转基因植株, 并且在田间表现出生长旺盛及抗叶片衰老的特性。
Agrobacterium tumefaciens mediated genetic transformation of five tomato cultivars by using the PSAG12-ipt chimeric gene constructed by the specific promoter PSAG12 and isopentenyltransferase (ipt) gene. The results obtained were Forty-four transformed plants from 31 different explants. Differentiation of infected cotyledons regeneration and transformation of plant growth in the field normal. Some of them transformed into regenerated plants by PCR test proved that transgenic plants, and in the field showed strong growth and anti-leaf senescence characteristics.
