目的 :探讨细胞凋亡与小肠粘膜萎缩发生之间的关系。材料和方法 :采用原位末端标记、免疫组化和 RT-PCR的方法 ,对正常小肠粘膜上皮和萎缩小肠粘膜上皮细胞凋亡的发生及其相关基因的表达进行对比研究。结果 :萎缩小肠粘膜上皮细胞凋亡的发生率明显高于正常小肠 ( P<0 .0 5) ;C- MYC在萎缩小肠绒毛顶部异常高表达 ;BAX和 BCL- 2呈弥散分布 ,BCL- 2在萎缩小肠粘膜上皮的表达显著低于正常小肠 ( P<0 .0 5) ,BAX的表达虽无显著性差异 ,但 BCL- 2与 BAX的比值明显降低。萎缩小肠粘膜上皮 bcl- 2的 m RNA表达较正常小肠明显降低 ,而 c-myc的 m RNA表达明显增高。结论 :小肠粘膜萎缩的发生与其细胞凋亡发生率增高有关 ,而细胞凋亡的增多与 bcl-2和 c- myc的异常表达有密切关系
Objective: To investigate the relationship between apoptosis and intestinal mucosal atrophy. MATERIALS AND METHODS: The apoptosis of normal small intestinal mucosa epithelium and atrophic small intestinal mucosa epithelial cells and their related gene expression were compared by in situ end-labeling, immunohistochemistry and RT-PCR. Results: The incidence of atrophic intestinal mucosal epithelial cell apoptosis was significantly higher than that of normal small intestine (P <0.05); C-MYC was abnormally highly expressed on the top of atrophic intestinal villi; BAX and BCL- The expression of BAX in atrophic small intestinal mucosa epithelium was significantly lower than that in normal small intestine (P <0.05), although the expression of BAX was not significantly different, the ratio of BCL-2 to BAX was significantly decreased. The m RNA expression of bcl-2 in atrophic small intestinal mucosa was significantly lower than that in normal small intestine, while the expression of c-myc m RNA was significantly increased. Conclusions: The occurrence of intestinal mucosa atrophy is related to the increased incidence of apoptosis, while the increased apoptosis is closely related to the abnormal expression of bcl-2 and c-myc
