目的将重组腺病毒介导的MDA-7/IL-24基因与阿霉素(ADM)联合治疗裸鼠肝癌,探讨基因治疗和化疗相结合治疗肿瘤的新方法。方法构建携带人MDA-7/IL-24的重组腺病毒载体(Ad-MDA-7),单独使用该载体或ADM以及两者联合治疗裸鼠肝癌模型,并进行组织学观察。结果成功构建重组腺病毒并实现体内表达,Ad-MDA-7+ADM联合治疗组裸鼠平均生存时间为(83.80±4.82)d,与其他3组比较生存期明显延长(P<0.01);Ad-MDA-7+ADM组裸鼠肝癌体积明显小于ADM或Ad-MDA-7组(P<0.01)。联合治疗组少见肿瘤细胞异型性,部分细胞核染色质边集。结论MDA-7/IL-24联合阿霉素对裸鼠人肝癌转移模型有明显的协同抗肿瘤作用。
OBJECTIVE: To investigate the combination of gene therapy and chemotherapy in the treatment of cancer in nude mice with recombinant adenovirus-mediated MDA-7 / IL-24 gene and adriamycin (ADM). Methods The recombinant adenovirus vector (Ad-MDA-7) carrying human MDA-7 / IL-24 was constructed and used alone or in combination with ADM to treat hepatoma in nude mice. Results The recombinant adenovirus was successfully constructed and expressed in vivo. The mean survival time of nude mice treated with Ad-MDA-7 + ADM was (83.80 ± 4.82) d, significantly longer than that of the other three groups (P <0.01). Ad The volume of liver cancer in MDA-7 + ADM group was significantly smaller than that in ADM or Ad-MDA-7 group (P <0.01). Combination therapy group rare tumor atypia, part of the nuclear chromatin edge set. Conclusion MDA-7 / IL-24 combined with doxorubicin has obvious synergistic anti-tumor effect on the metastasis model of human hepatocellular carcinoma in nude mice.
