目的:建立HPLC同时测定野葛花中鸢尾黄素-7-O-木糖葡萄糖苷、鸢尾苷、鸢尾苷元含量的方法。方法:采用Kromasil C18色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈-0.2%磷酸水溶液采用梯度洗脱,流速1.0 mL.min-1,检测波长265 nm,柱温35℃。结果:鸢尾黄素-7-O-木糖葡萄糖苷、鸢尾苷、鸢尾苷元的线性范围分别为0.14~1.40(r=0.999 4),0.20~2.02(r=0.999 3),0.10~1.04μg(r=0.999 1);平均回收率(n=6)分别为99.5%(RSD 1.8%),99.6%(RSD 1.7%),99.8%(RSD 1.9%)。结论:该方法简便、准确,重复性好,为野葛花药材的质量控制提供了实验依据。
OBJECTIVE: To establish a HPLC method for the simultaneous determination of tectorigenin-7-O-xyloglucoside, tectorigen and tectorigenin in Pueraria lobata. Methods: Kromasil C18 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase consisted of acetonitrile-0.2% phosphoric acid solution with a gradient of 1.0 mL · min-1. The detection wavelength was 265 nm and the column temperature was 35 ℃. RESULTS: The linear ranges of tectorigenin-7-O-xyloglucoside, tectorigenin and tectorigenin were 0.14-1.40 (r = 0.999 4), 0.20-2.02 (r = 0.999 3), 0.10-1.04 μg (r = 0.999 1). The average recoveries (n = 6) were 99.5% (RSD 1.8%), 99.6% (RSD 1.7%) and 99.8% (RSD 1.9%), respectively. Conclusion: The method is simple, accurate and reproducible. It provides an experimental basis for the quality control of Radix Puerariae.
