目的 研究高三尖杉酯碱 (homoharringtonine,HHT)对 HL- 6 0细胞端粒酶活性的影响及诱导凋亡作用。方法 采用端粒重复序列扩增法 -酶联免疫吸附试验检测了 HL- 6 0细胞的端粒酶活性变化 ;用细胞形态学观察、DNA琼脂糖凝胶电泳和 DNA片段原位末端标记法检测了细胞凋亡现象。结果 0 .0 0 5~ 0 .5 0 μg/ ml HHT处理 HL- 6 0细胞 0~ 48小时 ,HL- 6 0细胞端粒酶活性呈剂量依赖性和时间依赖性下降。同时 ,HL- 6 0细胞发生了凋亡。结论 HHT有效抑制 HL- 6 0细胞的端粒酶活性 ,诱导细胞凋亡。
Objective To investigate the effect of homoharringtonine (HHT) on telomerase activity and apoptosis in HL-60 cells. Methods The telomerase activity of HL-60 cells was detected by telomeric repeat amplification-enzyme-linked immunosorbent assay. Cell morphology, DNA agarose gel electrophoresis and DNA fragment in situ end-labeling Apoptosis phenomenon. Results The telomerase activity of HL-60 cells in a dose-dependent and time-dependent manner decreased from 0. 0 0 to 0.5 0 μg / ml HHT for 0-48 hours. At the same time, HL-60 cells developed apoptosis. Conclusion HHT can effectively inhibit telomerase activity and induce apoptosis in HL-60 cells.
