本文在60只蒙古沙土鼠研究了戊巴比妥对脑缺血的保护作用及对缺血性脑组织中单胺类神经介质含量的影响。动物随机分成三组:戊巴比妥组,对照组和模拟手术组各20只。戊巴比妥组和对照组分别于术前30分钟给予戊巴比妥钠60mg/kg体重和等容积的生理盐水1P,并作左侧颈总动脉结扎术。模拟手术组除不结扎左颈总动脉外,其它处理均同对照组。术后3小时检查动物的神经系统表现及测定脑组织中单胺类神经介质的含量。 发现戊巴比妥组动物卒中指数下降,神经系统症状明显改善,缺血性脑组织中去甲肾上腺素(NE),多巴胺(DA)和5-羟色胺升高至正常水平;动物的卒中指数与缺血性脑组织中NE、DA及5-HT的含量呈显著负相关(r分别=-0.485,-0.497和-0.579,P均<0.01)。动物的缺血症状越重,单胺类神经介质含量下降越明显。本文结果提示,戊巴比妥对脑缺血的保护作用可能与其改善缺血性脑组织中单胺类神经介质的代谢紊乱有关。
This paper studied the protective effect of pentobarbital on cerebral ischemia and the effect of monoamine neurotransmitters in ischemic brain tissue in 60 Mongolian gerbils. Animals were randomly divided into three groups: pentobarbital group, control group and simulated operation group of 20 animals each. In the pentobarbital group and the control group, sodium pentobarbital 60 mg/kg body weight and an equal volume of physiological saline 1P were administered 30 minutes before surgery, and left common carotid artery ligation was performed. In the simulated surgery group, except that the left common carotid artery was not ligated, the other treatments were the same as the control group. Neurological manifestations of the animals were examined 3 hours after surgery and the contents of monoamine neurotransmitters in brain tissue were determined. In the pentobarbital group, the index of stroke was decreased, neurological symptoms were significantly improved, and norepinephrine (NE), dopamine (DA), and serotonin increased to normal levels in the ischemic brain; animals had an index of stroke. There was a significant negative correlation between NE, DA, and 5-HT levels in ischemic brain tissue (r = -0.485, -0.497, and -0.579, respectively, P <0.01). The heavier the ischemic symptoms of animals, the more pronounced the decrease in monoamine neuronal content. The results of this study suggest that the protective effect of pentobarbital on cerebral ischemia may be related to the improvement of metabolic disorders of monoamine neurotransmitters in ischemic brain tissue.
