DNA甲基化和组蛋白乙酰化是表观遗传学经典的调节途径。其中,组蛋白乙酰化途径在脑胶质瘤发生过程中起到重要作用,并受组蛋白乙酰基转移酶(histone acetyltransferase,HAT)和组蛋白去乙酰化酶(histone deacetylase,HDAC)两类酶调节。组蛋白去乙酰化酶抑制剂(HDAC inhibitors,HDACIs)可以通过改变细胞内组蛋白乙酰化程度改变染色质空间结构,从而抑制目的基因转录,对肿瘤发生起到关键作用。本文就组蛋白乙酰化修饰与脑胶质瘤的关系及HDACIs在脑胶质瘤治疗中的应用进行综述。
DNA methylation and histone acetylation are classical epigenetic regulatory pathways. Among them, the histone acetylation pathway plays an important role in the pathogenesis of glioma and is regulated by histone acetyltransferase (HAT) and histone deacetylase (HDAC) adjust. Histone deacetylase inhibitors (HDACs) can change the spatial structure of chromatin by altering the degree of intracellular histone acetylation, thereby inhibiting the transcription of the target gene and playing a key role in tumorigenesis. This review summarizes the relationship between histone acetylation and glioma and the application of HDACIs in the treatment of glioma.
