目的分析广东省惠州市公共场所集中空调系统冷却水及冷凝水中分离的嗜肺军团菌的基因特征,初步建立惠州市嗜肺军团菌脉冲场凝胶电泳(PFGE)分型数据库。方法采用PFGE技术,对2012-2016年惠州市公共场所集中空调系统冷却水及冷凝水中分离的35株嗜肺军团菌进行电泳分析获得指纹图谱,并用BioN umerics 6.6软件对其进行聚类分析。结果 35株嗜肺军团菌分为29种不同的PFGE带型,菌株间带型相似系数为21.70%~100.00%。每种带型包含1~3株菌,存在5组带型一样的菌株,不存在优势型别;相同PFGE型别的菌株均来源于同一场所;同一场所存在2种以上不同PFGE型别;部分场所的冷却水和冷凝水中分离的菌株存在相同PFGE型别;相同血清型别的菌株经PFGE分型后得到不同的图谱,各菌株间PFGE图谱差异较大。结论惠州市公共场所集中空调冷却水及冷凝水中分离的嗜肺军团菌呈现基因多样性。
Objective To analyze the genetic characteristics of Legionella pneumophila isolated from the cooling water and condensate in the central air-conditioning system of public places in Huizhou City, Guangdong Province, and to establish the PFGE typing database of Legionella pneumophila in Huizhou City. Methods PFGE was used to analyze the fingerprint of 35 Legionella pneumophila isolated in the cooling water and condensate water of the central air-conditioning system in public places in Huizhou City from 2012 to 2016. The fingerprint was analyzed by BioNumerics 6.6 software. Results Thirty-five Legionella pneumophila strains were divided into 29 different PFGE patterns. The similarities of the bands between strains ranged from 21.70% to 100.00%. Each band type contains 1 to 3 strains, there are 5 strains with the same band type, there is no dominant type; the same PFGE type strains are from the same place; the same place there are more than two different PFGE types; part The same type of PFGE existed in the isolates of cooling water and condensate in the place. Different strains of the same serotypes obtained different maps after PFGE typing, and the PFGE patterns differed greatly among the isolates. Conclusion The genetic diversity of Legionella pneumophila isolated from the cooling water and condensate in central air-conditioning system in Huizhou City was variable.
