目的:研究大孔树脂纯化祖师麻中香豆素类成分的工艺。方法:以祖师麻甲素、总香豆素为指标,考察了3种大孔树脂的吸附效果并系统研究大孔树脂对祖师麻中香豆素类成分的吸附性能和洗脱参数。结果:D-101大孔树脂对祖师麻甲素、总香豆素有较好的纯化性能,最佳上样工艺是药液质量浓度生药50 g.L-1,上柱流速2 BV.h-1,pH 3.0。上样后先用4 BV水洗脱,继以5 BV50%乙醇洗脱,洗脱液流速2 BV.h-1。结论:确定的大孔树脂纯化祖师麻中的香豆素类成分的工艺是合理的。
Objective: To study the process of purification of coumarins from macroporous resins by using macroporous resin. Methods: The adsorption effects of macroporous resins on coumarin in Zujianma were systematically investigated using the macroporous resin as the index. Results: D-101 macroporous resin had better purification performance for Zhati ephedrine and total coumarin. The optimum loading condition was 50 gL-1 of crude drug concentration, 2 BV.h-1 , pH 3.0. After loading, the sample was eluted with 4 BV water followed by elution with 5 BV50% ethanol at a flow rate of 2 BV.h-1. CONCLUSIONS: The process of purifying coumarin components in the genus Zushi by macroporous resin is reasonable.
