给予大鼠ip MnCl_2·4H_2() 15mg/kg·d 84天抑制肝微粒体氨基比林N—脱甲基酶活性和细胞色素P—450含量下降,经ip PAS—Na 80mg/kg·d或12Omg/kg·d 21天后,氨基比林N—脱甲基酶活性和细胞色素P—450含量明显恢复。体外实验结果表示,锰时氨基比林N—脱甲基酶有直接抑制作用,而PAS—Na对该酶有直接激活作用。锰和PAS—Na对细胞色素P—450含量无明显直接效应。
Administration of ip MnCl 2 · 4H 2 (15 mg / kg · d) for 84 days decreased the activity of aminopyrine N-demethylase and the content of cytochrome P-450 in the liver microsomes. After ip PAS-Na 80 mg / kg · d or After 21 days of 12Omg / kg · d, aminopyrine N-demethylase activity and cytochrome P-450 content recovered significantly. In vitro results showed that aminopyrine N-demethylase had a direct inhibitory effect on manganese, while PAS-Na had a direct activation of the enzyme. Manganese and PAS-Na had no obvious direct effect on the content of cytochrome P-450.
