目的探讨DEHP对的小鼠的遗传毒性及氧化损伤。方法用不同剂量的邻苯二甲酸二异辛酯[di-(2-ethyl-hexyl)phthalate,DEHP]对小鼠每日腹腔注射染毒2周,通过彗星实验检测染毒2周小鼠的脑细胞DNA损伤,并测定肝,睾丸,脑组织中超氧化物歧化酶的活性变化。结果随着DEHP浓度增高,小鼠脑组织细胞DNA的损伤随之加重,DEHP的浓度达到125mg/kg和375mg/kg时,其可造成DNA的显著断裂(P<0.01)。肝、睾丸、脑的SOD活性变化是随DEHP浓度由低到高而先升高后降低,375mg/kg与对照组有显著性差异。结论DEHP可造成小鼠脑组织细胞DNA的损伤,及各脏器的氧化损伤。
Objective To investigate the genotoxicity and oxidative damage of DEHP-treated mice. Methods The mice were injected intraperitoneally with different doses of di- (2-ethyl-hexyl) phthalate (DEHP) for 2 weeks. The mice were exposed to comet assay for 2 weeks DNA damage of brain cells and the changes of superoxide dismutase activity in liver, testis and brain were measured. Results With the increase of DEHP concentration, the DNA damage in brain cells of mice was aggravated. At the concentrations of 125 mg / kg and 375 mg / kg DEHP, the DNA could cause significant DNA damage (P <0.01). The changes of SOD activity in liver, testis and brain were increased firstly and then decreased with the decreasing of DEHP concentration, while 375mg / kg had significant difference with the control group. Conclusion DEHP can cause DNA damage in mouse brain cells and oxidative damage in various organs.
