利用HPLC法研究了北五加皮不同时间水提物中绿原酸含量的变化规律。采用色谱柱Agilent ZORBAX SB-C18(150mm×4.6mm,5μm),流动相为0.4%磷酸溶液(A)-乙腈(B),梯度洗脱[0~8min 24%B;8~12min 50%B],检测波长278nm,流速为1.0mL/min,柱温为25℃。结果表明:绿原酸的最佳提取时间为2h,该方法简单快速、可行,含量测定准确。
The content of chlorogenic acid in aqueous extract of Pealwood and Peel at different times was studied by HPLC. The mobile phase consisted of a 0.4% phosphoric acid solution (A) -acetonitrile (B) with gradient elution [0-8 min 24% B; 8-12 min 50% B on a column Agilent ZORBAX SB-C18 ], Detection wavelength 278nm, flow rate 1.0mL / min, column temperature 25 ℃. The results showed that the optimum extraction time of chlorogenic acid was 2h. The method was simple, rapid, accurate and accurate.
